Химеры и мутанты ЦГТаз с прицелом на специфичность продукции γ-циклодекстрина

Аннотация:

За 135 лет изучения циклодекстринов и всего что с ними связано сделано очень много. Циклодекстрины представляют собой кольцевые молекулы из остатков глюкозы, образующиеся при ферментативной конверсии крахмала и формирующие тороидальные структуры с гидрофобной внутренней полостью и гидрофильной поверхностью, что позволяет им образовывать комплексы включения с целым рядом соединений. Поскольку с циклодекстринами неразрывно связаны бактериальные ферменты их продуцирующие, а именно циклодекстрин глюканотрансферазы (ЦГТазы), то можно считать, что 40 лет назад, когда были клонированы и секвенированы первые гены этих ферментов, начался новый молекулярно-биологический период их изучения. Это позволило выяснить доменную организацию ЦГТаз и выявить на аминокислотном уровне отличия между α-, β- и γ-специфичными ЦГТазами так как эти ферменты обычно продуцируют смесь разных циклодекстринов, состоящих из 6 (CD6), 7 (CD7) и 8 (CD8) остатков глюкопиранозы. При этом большинством ЦГТаз γ циклодекстрин образуется фактически «по остаточному принципу» и из-за этого в количественном отношении он сильно уступает своим «собратьям». Однако γ циклодекстрин представляет особый интерес, поскольку имеет внутреннюю полость большего размера и, следовательно может включать более крупные «гостевые» молекулы. С целью увеличить выход γ циклодекстрина созданы различные химеры и мутанты ЦГТаз, что явилось предметом рассмотрения в данной статье. Также уделено внимание прогнозам мирового производства циклодекстринов, включая γ циклодекстрин. Упомянуты и циклодекстрины с большим чем 8 числом остатков глюкозы.
