Конъюгаты полимерных β-циклодекстринов с сывороточным альбумином модулируют фармакокинетику и биораспределение антибиотика

Аннотация:

Синтезированы конъюгаты полимерного и мономер­ного гидроксипропил-β-циклодекстрина (ГПЦД) с человеческим сывороточным альбумином (ЧСА) как модельная система для модуляции фармакокинетики и биораспределения левофлоксацина (ЛВ). Структура синтезированных материалов подтверждена методом ИК-спектроскопии Фурье. Исследование кинетики высвобождения показало, что конъюгат ЧСА–ГПЦД обеспечивает наиболее выраженное замедление высвобождения препарата по сравнению со свобод­ным ЛВ. Исследования in vivo продемонстрировали существенное изменение тканевого распределения препарата. Использование конъюгатов увеличивало содержание ЛВ в легочной ткани с 5.4 до 8.7–9.7 мкг/г, что соответствует 1.6–1.8-кратному увеличению по сравнению со свободным препаратом. Одновременно концентрация антибиотика в печени снижалась с 11.2 до 6.5–8.9 мкг/г, а в почках — с 13.4 до 6.3–6.4 мкг/г, то есть в два раза. Кроме того, конъюгаты обеспечивали длительное поддержание высокой концентрации ЛВ в крови на протяжении всего периода наблюдения (480 мин). Полученные результаты свидетельствуют о перспективности данного подхода как механизма контроля биораспределения ЛВ за счет изменения сайта связывания антибиотика с ЧСА.
